What is Glutathione (GSH, Glutathione)?

This tripeptide helps cells protect themselves from damage by “scavenging” reactive species and maintaining the normal functioning of key processes in the body.

As a central element of redox homeostasis, GSH is the primary low-molecular-weight thiol in the cell and participates in Nrf2–Keap1 redox signaling and in enzyme-dependent pathways of the glutathione peroxidases/reductase (GPx/GR), which control the GSH/GSSG ratio and cellular oxidative status (summarized in Wikipedia: Glutathione).

Mechanism of action

In its reduced form (GSH) the molecule acts as an electron donor for GPx during peroxide neutralization, after which it is oxidized to GSSG and regenerated by GR with the participation of NADPH. It participates in conjugation reactions via glutathione-S-transferases (GST) and helps control protein S-glutathionylation. In erythrocytes and the liver GSH is a critical thiol-group buffer and indirectly influences cell signaling related to stress-response and detoxification (e.g., review articles in NCBI Bookshelf and publications indexed in PubMed).
